    An in-house contact at corporate told me there has been heavy talk regarding big changes happening in January. They didn't have specific details but believed that is why a national meeting is on hold. Someone had some sort of stress attack and crying at their desk about it last Friday? Odd

    “Why is the IV dosing regiment stretched over such a long period? Because Brexanolone is an endogenous steroid and therefore, tightly regulated.

    It has < 5% oral bioavailability, and is extensively metabolized by keto-reductase enzymes widely distributed in the body. The activity of these enzymes is itself subject to wild fluctuations from hormones, time since giving birth, weight, and individual genetic variability. By the way, for the person that mentioned that 9% of the patient "did not get the drug", I feel I need to educate you, even though simple logic is not what its used to be anymore. Its not that they did not "get the drug". They got it alright. It was just not detectable in plasma because their body metabolized it (degraded it for excretion in urine) so much that it did not reach a significant concentration in plasma (and therefore, cerebral fluids).

    The point is: the razor thin statistical significance observed for a sub-section (higher clinical depression) of a sub-section (detectable drug in plasma) of a sub-section (T = 0-12 days, not 15 days) will not increase with a larger patient population. And if you increase the dose too much to offset the biological machinery in place to regulate it, you will turn women into zombies.

    They did not know what to expect before starting this cie. They were right with the original hypothesis that slow IV dose would lead to clinical response. Then rain makers got involved, and reminded everyone that there would be no future without an oral dose. So they went for it under the general "self-absoluting" mantra that "science is an iterative process". Its also a highly lucrative process, even when failing up. Jonas is 20M$ richer for a drug treating 44 patients a quarter, and generating revenues 2 order of magnitudes lower then the operating cost.

    The next phase of that iterative process is weak to non-existing clinical results for the oral formulation, which is sort of settled science at this point.

    First quarter of 2020 is going to majorly suck. I do believe many will be putting in their notices in a matter of weeks as well as a reorg announcement is coming and jobs will be affected. My husband begged me to leave and I didn't listen. My new years resolution is to find a great job and get out by March. The working environment here never panned out as promised.

    Would you seriously go away. You are one person. Everyone knows it. It doesn't really matter. Facts are facts and this company is in the toilet and not making money. If you choose to stay, that is your problem.
